目的探讨地塞米松对哮喘豚鼠肺内、C7~T5脊神经节及对应节段脊髓背角内蛋白激酶C(PKC)表达的抑制作用。方法利用免疫组织化学和Western blotting方法,研究哮喘豚鼠肺内、C7~T5脊神经节及对应节段脊髓背角内PKC的免疫反应变化。结果哮喘组豚鼠肺内、C~T5脊神经节及对应节段脊髓背角内PKC平均光密度值明显高于对照组(P<0.01),而地塞米松组则明显低于哮喘组(P<0.01)。结论PKC可能参与哮喘的发病过程,地塞米松抑制哮喘豚鼠PKC的表达可能是其治疗哮喘的作用机制之一。
Objective To investigate the inhibitory effect of dexamethasone on the expression of protein kinase C (PKC) in the spinal dorsal horn of C7 ~ T5 and corresponding spinal segments in asthmatic guinea pigs. Methods Immunohistochemistry and Western blotting were used to study the changes of PKC in the dorsal horn of C7 ~ T5 spinal ganglia and corresponding spinal dorsal horn in asthmatic guinea pigs. Results The mean optical density of PKC in lungs of C - T5 spinal ganglia and corresponding segments of spinal cord in asthma group was significantly higher than that in control group (P <0.01), while the dexamethasone group was significantly lower than that in asthma group (P < 0.01). Conclusions PKC may be involved in the pathogenesis of asthma. Dexamethasone inhibits the expression of PKC in asthmatic guinea pigs and may be one of its mechanisms in the treatment of asthma.
