目的:探讨钙拈抗剂粉防己碱(tetradrine,Tet)对戊四唑(Pentylentetrazol,PTZ)诱导的大鼠癫癎模型的作用。方法:健康SD大鼠24只,随机分为4组,对照组和Tet 3个剂量组(15 mg/kg, 30 mg/kg,60 mg/kg)各6只。观察大鼠腹腔注射PTZ前后癫癎发作的情况,按Racine分级标准分级,同时记录皮层电图(ECoG),观察PTZ注射后到出现癎样放电的潜伏期及1 h内癎样放电累加的持续时间。电镜观察海马神经元超微结构的改变。结果:对照组给PTZ后均出现癫癎发作,程度均为5级, Tel组发作程度明显减轻。ECoG上出现癎样放电的潜伏期延长,癎样放电在1 h中的持续时间缩短。同时,Tel也能明显减轻PTZ致癎大鼠海马CA1区锥体细胞线粒体的损伤程度。结论:Tet对PTZ诱导的癫癎大鼠的发作有明显的对抗作用,对海马锥体神经元的损伤具有保护作用。
Objective: To investigate the effect of calcium antagonist tetradrine (Tet) on rat epileptic model induced by pentylenetetrazol (PTZ). Methods: Twenty-four healthy SD rats were randomly divided into 4 groups, 6 in control group and 3 in Tet group (15 mg / kg, 30 mg / kg, 60 mg / kg) The incidence of epileptic seizures before and after intraperitoneal injection of PTZ in rats was observed and graded according to the Racine grading standards. ECoG was also recorded. The latency of sublethal discharge after PTZ injection and the duration of sublethal discharge within 1 h were observed . Changes of Ultrastructure in Hippocampal Neurons Observed by Electron Microscopy. Results: Epileptic seizures occurred in both control and PTZ groups, all of which were grade 5 and significantly reduced in the Tel group. The ECoG appeared on the sample discharge extension of the incubation period, sample discharge in 1 h in the duration of shortening. At the same time, Tel also significantly reduced the mitochondrial damage of hippocampal CA1 pyramidal neurons in PTZ rats. CONCLUSION: Tet has a significant antagonistic effect on the onset of PTZ-induced epileptic rats and has a protective effect on pyramidal neurons in the hippocampus.
