目的建立保健食品中磷脂酰胆碱(PC)的高效液相色谱测定方法。方法样品用甲醇提取,采用高效液相色谱(HPLC)法测定PC的含量,色谱柱welch UltimateTMSiO2柱(5μm,250mm×4.6mm),流动相为甲醇,检测波长204nm,流速1.0mL/min,柱温30℃。结果 PC在0.0453~0.7248mg/mL范围内呈良好线性关系r=0.9999,最低检出限为0.325μg/mL,稳定性、重复性相对标准偏差<5%,平均回收率为98.9%。结论该法操作简便,结果准确,重现性好,分析速度快,检测灵敏度高,适用于卵磷脂类保健食品中PC含量的测定。
Objective To establish a method for the determination of phosphatidylcholine (PC) in health foods by high performance liquid chromatography. Methods The sample was extracted with methanol and the content of PC was determined by high performance liquid chromatography (HPLC). The column of welch Ultimate TMSiO2 (5μm, 250mm × 4.6mm) was eluted with a mobile phase of methanol at a detection wavelength of 204nm and a flow rate of 1.0mL / Temperature 30 ℃. Results PC showed good linearity in the range of 0.0453 ~ 0.7248mg / mL, with the lowest detection limit of 0.325μg / mL. The relative standard deviation of stability and repeatability was less than 5%, and the average recovery was 98.9%. Conclusion The method is simple, accurate, reproducible, fast in analysis and high in sensitivity. It is suitable for the determination of PC content in lecithin health food.
