目的:研究粉防己碱(Tet)对四氯化碳损伤的肝细胞的保护作用。方法:培养细胞中直接加入CCl_410mmol·L~(-1)造成损伤模型。膜流动性及胞内钙离子浓度分别用DPH,Fura 2-AM进行测定。结果:Tet(1-1000nmol·L~(-1))可以明显抑制CCl_4引起的肝细胞存活率降低和LDH的释放。结论:Tet具有保护肝细胞的作用。Tet的这种作用与其抑制肝细胞的脂质过氧化,稳定钙离子浓度,维持细胞膜流动性有关。
Objective: To study the protective effect of tetrandrine (Tet) on hepatocytes damaged by carbon tetrachloride. METHODS: Cultured cells were directly injected with CCl_410mmol·L -1 to induce the injury model. Membrane fluidity and intracellular calcium ion concentration were measured with DPH and Fura 2-AM, respectively. RESULTS: Tet (1-1000 nmol·L-1) significantly inhibited the decrease of hepatocyte survival rate and LDH release caused by CCl_4. Conclusion: Tet protects liver cells. This effect of Tet is related to its inhibition of lipid peroxidation of hepatocytes, stabilization of calcium ion concentration, and maintenance of cell membrane fluidity.
