目的:探讨高原植物芜菁挥发油成分对糖尿病小鼠动物模型的降血糖作用。方法:以超临界二氧化碳萃取仪提取芜菁挥发油成分。150只雄性小鼠采用高脂高糖喂养4周后,辅以连续3 d ip链尿佐菌素(STZ,100 mg·kg-1)造小鼠糖尿病模型,采用血糖测定仪尾静脉取血测空腹血糖,以血糖>11.0 mol·L-1为造模成功小鼠。将入选的40只小鼠随机分为正常对照组,模型组,芜菁挥发油高、低剂量(40,20 g·kg-1)组,阳性组(盐酸二甲双胍0.2 g·kg-1),共5组,ig每日1次,连续14 d,实验期间分别于造模前、造模后、用药后3,7,14 d剪尾取血检测血糖。实验结束后,小鼠摘眼球取血,取全血测定糖化血红蛋白,分离血清测定血清胰岛素水平,剖取新鲜的胸腺、脾脏,计算脏器指数,观察各组大鼠的血糖、脏器指数、糖化血红蛋白及胰岛素水平的变化。结果:芜菁挥发油成分可显著降低2型糖尿病模型小鼠的血糖(P<0.01),降低小鼠糖化血红蛋白(P<0.01,P<0.05),升高血清胰岛素水平(P<0.01)。结论:芜菁挥发油成分对STZ造成小鼠糖尿病模型有较明显的降血糖作用。
OBJECTIVE: To investigate the hypoglycemic effects of the essential oil of turnip in plateau on the animal model of diabetic mice. Methods: The volatile oil components of turnip were extracted by supercritical carbon dioxide extraction. 150 male mice were fed with high-fat and high-sucrose for 4 weeks and supplemented with streptozotocin (STZ, 100 mg · kg-1) for 3 days to make diabetic mice model. Blood was taken from the tail vein by blood glucose meter Fasting blood glucose was measured to blood glucose> 11.0 mol · L-1 for the modeling of successful mice. The selected 40 mice were randomly divided into normal control group, model group, high and low dose of turnip volatile oil (40,20 g · kg -1) group, positive group (metformin hydrochloride 0.2 g · kg -1) 5 groups, ig once daily for 14 days. During the experiment period, blood samples were taken before and 3, 7 and 14 days after the model making. At the end of the experiment, the mice were taken out of the eyeball, blood was taken from the eyeball, glycated hemoglobin was taken from the whole blood, the level of serum insulin was measured, the fresh thymus and spleen were separated and the organ index was calculated. The blood glucose, Glycated hemoglobin and insulin levels change. Results: The volatile oil components in turnip significantly reduced the blood glucose (P <0.01), reduced the HbA1c (P <0.01, P <0.05), and raised the level of serum insulin (P <0.01). Conclusion: The components of volatile oil of turnip have obvious hypoglycemic effect on STZ induced diabetic mice model.
