【目的】观察六味补气胶囊对慢性阻塞性肺病(COPD)大鼠T细胞介导转录调节因子(T-bet)、GATA-结合蛋白3(GATA-3)、孤独核受体(RORgammat)的影响。【方法】将50只SD大鼠随机分为正常组,模型组,六味补气高、中、低剂量组(剂量分别为0.7、0.35、0.175 g·kg-1·d-1),金水宝组(剂量为0.495 g·kg-1·d-1),脾氨肽组(剂量为0.33 mg·kg-1·d-1)。除正常组外,其他组大鼠均采用烟熏加脂多糖(LPS)气管滴入方法复制COPD模型,造模周期28 d后给药,连续给药30 d。采用动物肺功能仪检测大鼠肺功能,酶联免疫吸附法观察大鼠血清干扰素γ(IFN-γ)、白细胞介素(IL)-4、IL-17表达,Western blot法检测肺组织T-bet、GATA-3、RORgammat蛋白表达。【结果】与正常组比较,模型组大鼠肺功能值降低,血清IL-4和肺组织GATA-3表达降低,IFN-γ、IL-17、Th1/Th2升高,肺组织T-bet、T-bet/GATA-3、RORgammat表达升高(P<0.05或P<0.01)。与模型组比较,六味补气组肺功能值升高,IFN-γ、IL-17、Th1/Th2、T-bet、RORgammat表达降低,GATA-3表达升高(P<0.05或P<0.01)。【结论】六味补气胶囊通过上调GATA-3表达和下调T-bet、RORgammat表达而改善肺功能。
【Objective】 To observe the effects of Liuwei Buqi Capsule on the expression of T-bet, GATA-3 and RORgammat in chronic obstructive pulmonary disease (COPD) rats. influences. 【Methods】 Fifty Sprague-Dawley rats were randomly divided into normal group, model group, high, medium and low dose Liuwei Buqi (the dosage was 0.7,0.35,0.175 g · kg-1 · d-1) Group (dose of 0.495 g · kg-1 · d-1) and splenomegaly group (dose of 0.33 mg · kg-1 · d-1). Except for the normal group, COPD model was duplicated by tracheal instillation with lipopolysaccharide (LPS) by tracheal instillation method in rats of other groups. The model was given after 28 days and administered continuously for 30 days. The lung function of rats was detected by animal lung function test. The expression of IFN-γ, IL-4 and IL-17 were detected by enzyme-linked immunosorbent assay (ELISA) -bet, GATA-3, RORgammat protein expression. 【Results】 Compared with the normal group, the lung function of the model group decreased, the expression of IL-4 in serum and GATA-3 decreased, the levels of IFN-γ, IL-17 and Th1 / Th2 increased, T-bet, T-bet / GATA-3, RORgammat expression increased (P <0.05 or P <0.01). Compared with the model group, the lung function value of Liuweixuqi group was increased, the expression of IFN-γ, IL-17, Th1 / Th2, T-bet and RORgammat were decreased and the expression of GATA-3 was increased (P <0.05 or P <0.01) . 【Conclusion】 Liuwei Buqi Capsule can improve pulmonary function by up-regulating GATA-3 expression and down-regulating T-bet and RORgammat expression.
