目的 研究人骨肉瘤细胞凋亡及p5 3、p16、bcl- 2的表达情况 ,并初步探讨其临床及生物学意义。方法 采用原位DNA末端标记法及免疫组化方法检测了 39例骨肉瘤细胞凋亡及p5 3、p16、bcl- 2的表达情况 ,并统计其阳性反应率。结果 本组病例中细胞凋亡阳性率 6 1 5 % ,p5 3呈阳性反应 2 9例 ,阳性率 74 3% ,p16呈阳性反应 2 7例 ,阳性率 6 9 2 % ,bcl- 2呈阳性反应 7例 ,阳性率 19%。结论 骨肉瘤组织中有p5 3、p16、bcl- 2基因的高表达 ,与肿瘤的预后相关。在骨肉瘤中细胞凋亡是一种自然现象 ,其发生情况各不相同 ,研究初步提示了其与预后相关 ,但应与p5 3、p16、bcl- 2蛋白的表达相结合而综合判断。
Objective To study the apoptosis of human osteosarcoma cells and the expression of p53, p16 and bcl-2, and to discuss their clinical and biological significance. Methods The expression of p53, p16 and bcl-2 was detected in 39 cases of osteosarcoma cells by in situ DNA end labeling and immunohistochemistry. The positive reaction rate was counted. Results The positive rate of apoptosis was 61.5 % in this group of patients, p53 was positive in 29 cases, the positive rate was 74 3%, p16 was positive in 27 cases, the positive rate was 692%, and bcl-2 was positive. The response was 7 cases and the positive rate was 19%. Conclusion The high expression of p53, p16 and bcl-2 gene in osteosarcoma is related to the prognosis of tumors. Apoptosis is a natural phenomenon in osteosarcoma, and its occurrence is not the same. The study initially suggests that it is associated with prognosis, but should be combined with the expression of p53, p16, bcl-2 protein and comprehensive judgment.
