在进行甾体、核苷酸、多肽以及甘油酯等多功能复杂分子的反应与合成时,除特定部位基团发生预期反应外,还常常导致其它基团的变化,结果使反应和产物极其复杂。为使反应按预期方向进行,需对甾体化合物等分子中一些活性基团加以保护,待反应完毕,再解除保护基。显然,选用的保护基应具有一定的稳定性,能经受反应条件;解除保护基时,应能再生原有基团,又不影响分子的其它部分;
