目的 了解不同转移能力的涎腺腺样囊性癌 (salivaryadenoidcysticcarcinoma ,SACC)细胞与细胞外基质粘附、对细胞外基质侵袭及趋化运动的能力。方法 MTT法测定SACC -LM ,SACC - 83,ACC -M ,ACC - 2与纤维粘连蛋白、层粘连蛋白、Ⅳ型胶原的粘附 ,用改良Boydenchamber方法观察了SACC细胞对人工重组基底膜的侵袭以及趋化运动。结果 高转移的SACC -LM与细胞外基质的粘附低于SACC - 83,而ACC -M与细胞外基质的粘附高于ACC - 2。SACC与纤维粘连蛋白的粘附高于和层粘连蛋白、Ⅳ型胶原的粘附。SACC -LM ,ACC -M的侵袭和运动能力强于SACC - 83,ACC - 2。结论 不同转移能力的SACC细胞与细胞外基质的粘附能力不同。高转移潜能的涎腺腺样囊性癌细胞的侵袭能力、趋化运动能力较强
Objective To investigate the ability of SACC cells with different metastatic potential to adhere to extracellular matrix and to extracellular matrix invasion and chemotaxis. Methods The adhesion of SACC - LM, SACC - 83, ACC - M and ACC - 2 to fibronectin, laminin and type Ⅳ collagen were measured by MTT assay. The invasion of artificial recombinant basement membrane was observed by modified Boydenchamber method As well as chemotaxis. Results The adhesion of highly metastatic SACC - LM to extracellular matrix was lower than that of SACC - 83, whereas ACC - M adhered to extracellular matrix more than ACC - 2. Adhesion of SACC to fibronectin was higher than that of laminin and type IV collagen. SACC-LM and ACC-M had stronger invasion and exercise capacity than SACC-83 and ACC-2. Conclusion SACC cells with different metastatic potential have different adhesion ability to extracellular matrix.
