用重组人粒细胞-巨噬细胞集落刺激因子-白细胞介素3(rbGM-CSF-IL3)融合蛋白(G3)作为免疫原.通过B淋巴细胞杂交瘤技术建立了两株稳定分泌抗rhGM-CSF-IL3(G3)单克隆抗体的杂交瘤细胞株.分别命名为MG31和MG32。经鉴定,两种单抗均为小鼠IgG1亚类。其染色体众数分别为91和86.并能特异地识别分子量约30KD的G3。MG31和MG32经3个月传代培养及冻存复苏.在培养沿及Balb/c小鼠腹水中均能产生而滴度抗体。抗G3单克隆抗体的获得为深入研究G3的亲和层析纯化、G3的免疫学测定及其生物学特性提供了必要的条件。
Recombinant human granulocyte-macrophage colony stimulating factor-interleukin-3 (rbGM-CSF-IL3) fusion protein (G3) was used as an immunogen. Two hybridoma cell lines stably secreting anti-rhGM-CSF-IL3 (G3) monoclonal antibody were established by B lymphocyte hybridoma technique. Named MG31 and MG32 respectively. Identified, the two monoclonal antibodies are mouse IgG1 subclass. The chromosomes were 91 and 86 respectively. And can specifically identify G3 with a molecular weight of about 30KD. MG31 and MG32 after 3 months subculture and cryopreservation. Antibodies were titrated in both ascites and Balb / c mice. The obtaining of anti-G3 monoclonal antibody provided necessary conditions for in-depth study on purification of G3 by affinity chromatography, immunological determination of G3 and its biological characteristics.
