目的:观察慢性低氧对大鼠肺组织、肺动脉组织及大鼠远端肺动脉平滑肌细胞(PASMCs)中骨形态发生蛋白4(BMP4)的内源性拮抗蛋白基因Chordin mRNA表达的影响。方法:12只雄性SD大鼠完全随机分为2组(n=6):即常氧对照组和低氧21 d组;原代培养大鼠远端PASMCs,分为2组(n=6):即常氧对照组和低氧60 h组,采用实时定量PCR检测Chordin mRNA表达水平。结果:1低氧21 d大鼠与常氧状态下的大鼠相比较,在肺组织、肺动脉组织及肺动脉平滑肌内的Chordin的mRNA的表达均有增加,分别是增加1.36倍、1.24倍、1.58倍(P<0.05);2低氧60 h的大鼠远端PASMCs,与对照组细胞比较,BMP内源性拮抗蛋白Chordin的mRNA的表达增加1.79倍(P<0.05)。结论:BMP4内源性拮抗蛋白Chordind mRNA表达在慢性低氧初期对抗增加的BMP4表达增加,对于CHPH发病可能具有保护作用。
Objective: To observe the effects of chronic hypoxia on the expression of endogenous antagonist protein Chordin mRNA of bone morphogenetic protein 4 (BMP4) in rat pulmonary tissue, pulmonary artery and rat distal pulmonary artery smooth muscle cells (PASMCs). Methods: Twelve male Sprague-Dawley rats were randomly divided into 2 groups (n = 6): normoxia control group and hypoxia for 21 days. Primary cultured rat PASMCs were divided into 2 groups (n = 6) : Normoxic control group and hypoxia 60 h group, using real-time quantitative PCR to detect Chordin mRNA expression levels. Results: Compared with rats in normoxia state, the expression of Chordin mRNA in lung tissue, pulmonary artery and pulmonary artery smooth muscle of hypoxic 21 d rats increased by 1.36, 1.24 and 1.58 (P <0.05) .2 Compared with the control cells, the mRNA expression of Chordin, an endogenous BMP protein, increased by 1.79-fold in the distal PASMCs exposed to hypoxia 60 h (P <0.05). CONCLUSION: Chordind mRNA, an endogenous antagonist of BMP4, may antagonize the increase of BMP4 expression in the early stage of chronic hypoxia and may have a protective effect on the pathogenesis of CHPH.
