目的:探讨痹祺胶囊对胶原诱导性关节炎(CIA)大鼠骨桥蛋白(OPN)表达的影响。方法:建立CIA大鼠模型,检测OPN在血清、滑膜、软骨中的表达及血清TNF-α水平,观察软骨病理变化。结果:模型组较正常组大鼠OPN在血清、滑膜及软骨表达量及血清TNF-α水平升高(P<0.05),各治疗组较模型组表达量降低(P<0.05);模型组较正常组软骨损伤明显(P<0.05),各治疗组较模型组软骨损伤明显改善(P<0.05)。结论:初步证明OPN、TNF-α参与CIA的病理过程,痹祺胶囊治疗RA机制可能与其降低OPN表达及血清TNF-α水平有关,进而抑制RA的软骨破坏。
Objective: To investigate the effect of Biqi capsule on osteopontin (OPN) expression in rats with collagen induced arthritis (CIA). Methods: The CIA rat model was established. The expression of OPN in serum, synovial membrane and cartilage, serum TNF-α level and the pathological changes of cartilage were observed. Results: Compared with normal group, the expression of OPN in serum, synovium and cartilage and the level of serum TNF-α in model group were significantly increased (P <0.05), and the expression of OPN in model group was lower than that in model group (P <0.05) Compared with the normal group, the cartilage damage was significantly (P <0.05), and the cartilage damage in each treatment group was significantly improved (P <0.05). CONCLUSION: It is preliminary evidence that OPN and TNF-α are involved in the pathological process of CIA. The mechanism of action of Biqi capsule on RA may be related to the decrease of OPN expression and the level of serum TNF-α, thereby inhibiting the cartilage destruction of RA.
