目的:进行体外透皮吸收试验,考察磷脂固体分散技术对盐酸小檗碱体外透皮能力的影响。方法:以离体小鼠腹部皮肤为透皮屏障,采用改良Franz扩散池装置进行透皮吸收实验,采用紫外分光光度法测定透皮接收液中小檗碱含量,比较盐酸小檗碱及其磷脂固体分散体透皮能力的差异,评价磷脂固体分散技术对小檗碱透皮吸收的促进作用。结果:高、中、低浓度盐酸小檗碱磷脂固体分散体的累计透皮量均远高于同等浓度盐酸小檗碱。结论:磷脂固体分散技术可提高盐酸小檗碱的透皮能力,盐酸小檗碱磷脂固体分散体可用于透皮吸收制剂的研发。
OBJECTIVE: To study the transdermal absorption in vitro and investigate the effect of solid dispersion of phospholipid on the in vitro dermal penetration of berberine hydrochloride. Methods: The transdermal absorption of the mouse skin was used as the transdermal barrier of the isolated mouse skin. The percutaneous absorption of the transdermal solution was measured by the improved Franz diffusion cell. The content of berberine in the transdermal receiving fluid was determined by UV spectrophotometry. The effects of berberine hydrochloride and its phospholipid solid Dispersion of transdermal ability differences to evaluate the solid dispersion of phospholipid technology on the promotion of berberine transdermal absorption. Results: The cumulative transdermal volume of solid dispersion of berberine hydrochloride in high, middle and low concentrations was much higher than that of the same concentration of berberine hydrochloride. Conclusion: The solid dispersion of phospholipid can improve the transdermal ability of berberine hydrochloride. The solid dispersion of berberine hydrochloride can be used for the development of transdermal absorption preparations.
