目的研究尿激酶型纤溶酶原激活受体(uPAR)在肝癌组织及细胞系中的表达,探讨其与肝细胞癌侵袭、转移的关系。方法利用免疫组化方法检测50例肝癌组织中uPAR的表达,分析其与肝癌的临床病理分级、包膜侵犯、门脉癌栓等的关系;利用流式细胞术检测肝癌细胞系Hep-3B和SMMC-7721中uPAR的表达,通过Boyden小室侵袭实验比较两种细胞系体外侵袭能力的差别;从SMMC-7721中通过流式细胞术分选出uPAR阳性及阴性的细胞,比较两者体外侵袭能力的差别,并通过裸鼠体内成瘤及转移模型,比较两者体内侵袭和转移的能力。结果肝癌中uPAR的表达和肿瘤的包膜侵犯、门脉癌栓有显著的相关性(P<0.01);肝癌细胞系Hep-3B和SMMC-7721中uPAR的表达和其体外侵袭能力相关;而在SMMC-7721中,uPAR阳性组较阴性组有更强的体外及体内侵袭及转移能力。结论uPAR蛋白的表达在肝癌细胞的侵袭和转移中起重要作用,和肿瘤的临床病理分期及预后相关。
Objective To investigate the expression of urokinase-type plasminogen activator receptor (uPAR) in hepatocellular carcinoma (HCC) tissues and cell lines and to explore its relationship with invasion and metastasis of hepatocellular carcinoma. Methods The expression of uPAR in 50 HCC tissues was detected by immunohistochemistry. The relationship between the expression of uPAR and the clinicopathological grade, invasion of capsule and portal vein tumor thrombus in HCC was analyzed. Hep-3B and HCC were detected by flow cytometry The expression of uPAR in SMMC-7721 cells was compared by Boyden chamber invasion assay to compare the invasive ability of the two cell lines in vitro; uPAR positive and negative cells were sorted by flow cytometry from SMMC-7721, The nude mice in vivo tumorigenicity and metastasis model, the two in vivo invasion and metastasis ability. Results The expression of uPAR in hepatocellular carcinoma was significantly correlated with tumor invasion and portal vein tumor thrombus (P <0.01). The expression of uPAR in hepatocellular carcinoma cell lines Hep-3B and SMMC-7721 was related to its invasion ability in vitro. In SMMC-7721, uPAR positive group had stronger in vitro and in vivo invasion and metastasis than negative group. Conclusion The expression of uPAR protein plays an important role in the invasion and metastasis of hepatocellular carcinoma cells, and is related to the clinicopathological stage and prognosis of the tumor.
