目的 观察双黄连抗柯萨奇B3 病毒感染原代大鼠心肌细胞的作用 ,并初步探讨其机制。方法 选用出生 1~ 3天的Sprague Dawley大鼠 ,取其心室肌组织 ,制备原代大鼠心肌细胞 ,培养 72h后 ,用不同浓度的双黄连对抗10 0TCID50 柯萨奇B3 型病毒感染细胞 ,观察细胞病变 ,测定感染 36h后细胞上清液中的病毒滴度及肌酸磷酸激酶。结果 0 5、0 2 5、0 12 5 g/L双黄连均可明显减轻心肌细胞病变 ,降低细胞上清液的病毒滴度 ,减少感染心肌细胞肌酸磷酸激酶的释放 (分别和病毒对照组相比 ,P <0 0 1)。结论 双黄连对柯萨奇病毒感染的新生大鼠原代心肌细胞有保护作用。
Objective To observe the effect of Shuanghuanglian against Coxsackie B3 virus in infecting primary rat cardiomyocytes and to explore its mechanism. Methods Sprague Dawley rats aged 1 to 3 days were selected and their ventricular myocardium was taken to prepare primary rat cardiomyocytes. After 72 hours of culture, Shuanghuanglian was used to challenge 10 0TCID50 Coxsackie B3 virus infected cells. The cytopathic effect was determined by measuring the virus titer and creatine phosphokinase in the cell supernatant 36 h after infection. RESULTS: Shuanghuanglian, 0 5, 0 2 5, and 0 12 5 g/L all significantly attenuated cardiomyocyte pathological changes, reduced viral titer of cell supernatant, and decreased release of creatine kinase from infected cardiomyocytes (respectively, viral control group). In contrast, P <0 0 1). Conclusion Shuanghuanglian has protective effects on primary cardiomyocytes of newborn rats infected by Coxsackie virus.
