研究两个反义核酸(18mer,21mer)对人白血病HL—60细胞中c-myc基因表达的抑制作用。用HL-60活细胞计数,RNA含量的RT-PCR测定和c-myc基因蛋白的免疫组化染色检查。结果:两个反义寡核苷酸都能抑制HL-60细胞的增殖,增殖抑制率为11%~75%。反义核酸还抑制c-mycmRNA和Myc蛋白的表达,而对c-mcy基因正常表达的红白血病细胞K562的增殖和PHA刺激的人淋巴细胞的增殖没有影响。结论:反义寡核苷酸能抑制c-myc基因的过度表达。
The inhibitory effect of two antisense nucleic acids (18mer, 21mer) on c-myc gene expression in human leukemia HL-60 cells was investigated. HL-60 viable cell counts, RT-PCR assays of RNA content, and immunohistochemical staining of c-myc gene protein were examined. RESULTS: Both antisense oligonucleotides inhibited the proliferation of HL-60 cells and the proliferation inhibition rate was 11% to 75%. Antisense nucleic acids also inhibit the expression of c-myc mRNA and Myc protein, but have no effect on the proliferation of erythroleukemia cell K562 and the proliferation of PHA-stimulated human lymphocytes that are normally expressed in the c-mcy gene. Conclusion: Antisense oligonucleotides can inhibit the overexpression of c-myc gene.
