目的观察紫铆花素对人黑素瘤细胞A375细胞增殖、细胞周期、酪氨酸酶活性、黑素合成及线粒体膜电位的影响。方法用不同浓度紫铆花素处理人黑素瘤细胞A375 48h后,采用MTT法测定黑素细胞增殖率;流式细胞仪测定黑素细胞周期和线粒体膜电位变化;L-DOPA法测定黑素细胞酪氨酸酶活性及Na OH裂解法测定黑素含量变化。结果与对照组相比,0.1~5.0μg/m L浓度范围紫铆花素与人黑素瘤细胞A375共孵育48h后,能明显促进人黑素瘤细胞A375增殖,具有不同程度提高酪氨酸酶活性、促进黑素合成及调节细胞周期和线粒体膜电位的作用。结论紫铆花素对体外培养的人黑素瘤细胞A375起促进细胞增殖、提高酪氨酸酶活性、增加黑素合成的作用,其作用可能与紫铆花素对细胞周期和线粒体膜电位的调节有关。
Objective To investigate the effects of Variscariae on proliferation, cell cycle, tyrosinase activity, melanin synthesis and mitochondrial membrane potential of human melanoma A375 cells. Methods Human melanoma A375 cells were treated with different concentration of rivalanin for 48 hours. The proliferation rate of melanocytes was measured by MTT assay.
