建立了高效液相色谱法测定不同产地补骨脂中的6种呋喃香豆素化合物:补骨脂素、8-甲氧基补骨脂素、异补骨脂素、5-甲氧基补骨脂素、欧前胡素和三甲基补骨脂素。采用Capcell Pak-C_(18)色谱柱,以甲醇∶水为流动相,线性梯度洗脱,检测波长248 nm。结果 6种成分的峰面积在1~100μg/ml范围内线性关系良好,平均加样回收率分别为97.2%、99.5%、96.8%、99.2%、99.6%和98.7%,RSD分别为1.54%、1.35%、0.65%、0.43%、0.18%和0.26%。10种不同产地补骨脂中上述6种成分的含量分别为0.21%~0.86%、0.10%~0.17%、0.28%~0.89%、0.14%~0.43%、0.10%~0.21%和0.10%~0.26%。经方法学验证,本方法可用于补骨脂药材的质量控制。
A HPLC method was established for the determination of six furanocoumarins in psoralen of different origins: psoralen, 8-methoxy psoralen, isopsoralen, 5-methoxyacid Psoralen, imperatorin and trimethyl psoralen. Capcell Pak-C_ (18) column was used with a linear gradient of methanol: water as the mobile phase. The detection wavelength was 248 nm. Results The peak areas of the six components ranged from 1 μg / ml to 100 μg / ml. The average recoveries were 97.2%, 99.5%, 96.8%, 99.2%, 99.6% and 98.7%, respectively, with RSDs of 1.54% 1.35%, 0.65%, 0.43%, 0.18% and 0.26%. The contents of the above six components in 10 different producing areas of Psoralen were respectively from 0.21% to 0.86%, from 0.10% to 0.17%, from 0.28% to 0.89%, from 0.14% to 0.43%, from 0.10% to 0.21% and from 0.10% to 0.26 %. The methodological verification, the method can be used for the quality control of psoralen herbs.
