目的探讨kdr基因在家蝇溴氰菊酯抗性增加中的作用。方法首次将高分辨率熔解曲线(high resolution melting,HRM)分析与半定量PCR相结合,用于分析家蝇抗性基因kdr扩增和突变情况。对扩增的PCR产物进行测序来验证HRM准确性。结果与敏感品系家蝇相比,kdr的mRNA表达量在现场品系家蝇中没有显著改变(P>0.05)。使用HRM对kdr进行突变分析,发现所有样本均为同一基因型。经核酸测序,该扩增片段没有突变位点。结论杭州家蝇溴氰菊酯抗性增加与kdr基因无关。
Objective To investigate the role of kdr gene in deltamethrin resistance in housefly. Methods For the first time, high resolution melting (HRM) analysis combined with semi-quantitative PCR was used to analyze the amplification and mutation of the kdr gene in housefly. The amplified PCR product was sequenced to verify HRM accuracy. Results Compared with the susceptible housefly, the expression of kdr mRNA did not change significantly in the housefly (P> 0.05). Mutation analysis of kdr using HRM revealed that all samples were of the same genotype. After nucleic acid sequencing, there was no mutation in the amplified fragment. Conclusion The increase of deltamethrin resistance in housefly of Hangzhou has nothing to do with kdr gene.
