目的:探讨HLA-DQβ链第57位非门冬氨酸(NA)或门冬氨酸(A)与青少年1型糖尿病易感性或抵抗性的关系。方法:采用PCR-SSOP法对10例1型糖尿病病人和20例健康对照者进行HLA-DQβ1基因分型。结果:NA和A频率在1型糖尿病组分别为61.7%和38.5%,在对照组中分别为38.4%和60.6%(P<0.01);表现型NA/NA、NA/A和A/A频率在1型糖尿病组中分别为46.5%、34.6%和19.1%,在对照组中分别为30.2%、16.4%和52.6%,其中表现型A/A频率在1型糖尿病组的分布明显较对照组降低(P<0.01)。结论:研究提示NA与青少年1型糖尿病的易感性呈正相关,A与青少年1型糖尿病的易感性呈负相关。
Objective: To investigate the relationship between the 57th non-aspartic acid (NA) or aspartic acid (A) in HLA-DQβ chain and the susceptibility or resistance to type 1 diabetes in adolescents. Methods: HLA-DQβ1 genotyping was performed on 10 type 1 diabetic patients and 20 healthy controls by PCR-SSOP. Results: The frequencies of NA and A were 61.7% and 38.5% in type 1 diabetic patients and 38.4% and 60.6% (P <0.01) in control subjects respectively. The frequencies of NA / NA, NA / A and A / A 46.5%, 34.6% and 19.1% in type 1 diabetes mellitus group, and 30.2%, 16.4% and 52.6% in control group, respectively. The distribution of phenotype A / A frequency in type 1 diabetes mellitus group was significantly higher than that in control group Decreased (P <0.01). Conclusion: This study suggests that NA has a positive correlation with susceptibility to type 1 diabetes in adolescents, and that A has a negative correlation with susceptibility to type 1 diabetes in adolescents.
