①目的探讨体外授精(IVF)及胚胎移植(ET)工作中培养液准备与早期胚胎发育的关系。②方法分别检测普通法、微滴法和人脐血清培养液中鼠正常形态胚泡形成率,并进行统计学分析。③结果单纯Ham'sF10培养液(普通法,Ⅰ组)中正常形态胚泡形成率为35.89%,而微滴法(Ⅱ组)的胚泡形成率为74.01%,两者相比差异具有极显著意义(χ2=90.282,P<0.001);用微滴法准备培养液时,含有10%人灭活脐血清的Ham'sF10中(Ⅲ组),胚泡发育率为68.02%,与第Ⅱ组比较无显著差异(χ2=2.971,P>0.05)。④结论液状石蜡油具有稳定微培养环培作用;人脐血清无改善早期鼠胚发育的作用
Objective To explore the relationship between preparation of culture medium and development of early embryos during in vitro fertilization (IVF) and embryo transfer (ET). Methods The common normal, microdialysis method and human umbilical cord blood serum culture medium were used to detect the normal blastocyst formation rate and analyzed statistically. Results The formation rate of blastocyst in normal Ham’sF10 culture medium (common method, group Ⅰ) was 35.89%, while the droplet method (group Ⅱ) blastocyst formation rate was 74.01% (Χ2 = 90.282, P <0.001). In the preparation of culture solution by microdrop method, blastocyst development was observed in Ham’sF10 containing 10% human inactivated cord serum (group Ⅲ) The rate was 68.02%, no significant difference compared with the second group (χ2 = 2.971, P> 0.05). Conclusion liquid paraffin oil has a stable micro-culture of cyclosporine; human umbilical serum did not improve early embryonic development
