目的 了解23个型的肺炎球菌荚膜多糖的稳定性。方法 分别用Sepharose2B或4B凝胶柱测定 相对分子质量及其分布(Kd)。结果 用蓝色葡聚糖2000和氧化钠作标准测定外水体积Vo(152.56或154.04min)和 内水体积Vt(432.23或444.07min),均有良好的分离效果。于2~8℃保存的所有型别的多糖均符合规程要求,除1、 2、4、8、12F、14、18C和23F型多糖因基础相对分子质量较大有轻微降解外,其他均未发现有明显降解;于室温保存的 多糖除1、2、8、12F、14和18C型有轻微降解外,其他型多糖降解程度均较大;于37℃保存的多糖除1、2、8、12F和14 型有轻微降解外,其他型则有更大程度的降解。结论 高温不利于多糖的保存。23个型别的液态多糖于2~8℃保 存1年均稳定。
Objective To understand the stability of 23 pneumococcal capsular polysaccharides. Methods The relative molecular mass and its distribution (Kd) were determined by Sepharose 2B or 4B gel column respectively. Results The external water volume Vo (152.56 or 154.04 min) and the internal water volume Vt (432.23 or 444.07 min) were determined with blue dextran 2000 and sodium oxide as standard, which showed good separation effect.
