目的:研究中药益气解毒方对大鼠局灶性脑缺血后氧化应激损伤的影响。方法:取健康雄性SD大鼠60只,随机分为6组:假手术组、模型组、阳性药银杏叶提取物组(4 mg.kg-1)、益气解毒高、中、低剂量组(25,5,1 mg.kg-1)。给药组于手术麻醉前10 min灌胃给药,用线栓法制作大鼠大脑中动脉闭塞(middle cerebral artery occlusion,MCAO)模型,12 h后取血清和缺血脑组织制备组织匀浆液,用酶标仪定量检测超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)、单胺氧化酶(MAO)、髓过氧化物酶(MPO)。结果:与假手术组比较,模型组脑组织SOD和GSH-Px活性明显降低(P<0.01),MDA含量和MAO,MPO活性明显升高(P<0.05)。与模型组比较,益气解毒方高、中、低剂量组、阳性药组SOD和GSH-Px显著性升高(P<0.01),MAO,MDA和MPO显著性降低(P<0.05,P<0.01)。结论:益气解毒方对大鼠局灶性脑缺血有保护作用,其作用机制可能与抗氧化应激作用有关。
Objective: To study the effect of Yiqi Jiedu Fang on oxidative stress injury after focal cerebral ischemia in rats. Methods: Sixty healthy male Sprague-Dawley rats were randomly divided into 6 groups: sham-operation group, model group, Ginkgo biloba extract group (4 mg.kg-1), Yiqi detoxification high, medium and low dose group (25, 5, 1 mg.kg-1). The rats in the treatment group were intragastrically administrated 10 min before the operation, and the middle cerebral artery occlusion (MCAO) model was established by the method of thread occlusion. After 12 h, the homogenate was prepared from the serum and the ischemic brain tissue, Quantitative detection of superoxide dismutase (SOD), malondialdehyde (MDA), glutathione peroxidase (GSH-Px), monoamine oxidase (MAO) and myeloperoxidase (MPO) by microplate reader. Results: The activities of SOD and GSH-Px in the model group were significantly lower than those in the sham operation group (P <0.01). The content of MDA and the activities of MAO and MPO in the model group were significantly increased (P <0.05). Compared with the model group, the SOD and GSH-Px in the high, middle and low doses of Yiqi Jiedu prescription group and the positive control group were significantly increased (P <0.01), and MAO, MDA and MPO were significantly decreased (P <0.05, P < 0.01). Conclusion: Yiqi Jiedu Fang has a protective effect on focal cerebral ischemia in rats, and its mechanism may be related to anti-oxidative stress.
