目的:观察参附汤对移植后小鼠造血干细胞归巢的影响。方法:以转GFP基因的Balb/c荧光小鼠作为供鼠,以普通Balb/c小鼠为受鼠,进行造血干细胞移植,移植后抽取4只小鼠作为单纯照射组以观察照射剂量是否充足外,其余小鼠随机分为参附组和空白组,参附组给予参附汤,空白组给予0.9%生理盐水灌胃,分别于移植后+3、+7、+14天随机抽取骨髓标本,用激光共聚焦显微镜观察荧光细胞的归巢数量,流式细胞仪检测骨髓单个核细胞的sca-1+细胞的表达率。结果:激光共聚焦显微镜检测显示,参附组小鼠造血干细胞归巢的数量较空白组增多,并随移植时间的推移,呈逐渐增多的趋势。流式检测显示,骨髓单个核细胞的sca-1+细胞+3、+7、+14天的表达率分别为(5.36±1.27)%、(5.77±1.14)%、(5.42±1.14)%,与空白组相比,二者间有显著性差异(P<0.05)。结论:具有温阳补肾作用的参附汤能促进自体移植小鼠造血干细胞的归巢。
Objective: To observe the effect of Shenfu Decoction on homing of hematopoietic stem cells in mice after transplantation. METHODS: Balb/c mice with GFP gene transfer were used as donors and normal Balb/c mice were used as recipients. Hematopoietic stem cell transplantation was performed. After transplantation, 4 mice were taken as simple irradiation group to observe whether the irradiation dose was sufficient. In addition, the remaining mice were randomly divided into Shenfu group and blank group. Shenfu group was given Shenfu decoction. The blank group was given 0.9% saline, and bone marrow samples were taken at +3, +7, +14 days after transplantation. The number of fluorescent cells homing was observed by laser confocal microscopy, and the expression rate of sca-1+ cells in bone marrow mononuclear cells was measured by flow cytometry. RESULTS: Laser confocal microscopy showed that the number of hematopoietic stem cells homing in the Shenfu group was greater than that of the blank group, and it gradually increased with the time of transplantation. Flow cytometry showed that the expression rates of sca-1+ cells in bone marrow mononuclear cells at days +3, +7, and +14 were (5.36±1.27)%, (5.77±1.14)%, (5.42±1.14)%, respectively. Compared with the blank group, there was a significant difference between the two groups (P<0.05). Conclusion: Shenfu decoction with Wenyang Bushen can promote the homing of hematopoietic stem cells in autograft mice.
