目的注释并分类红花Carthamus tinctorius中转录因子家族,分析其在不同组织间的表达水平差异。方法使用红花不同组织转录组测序数据组装Unigene,利用Nr数据库中转录因子信息比对并注释红花转录因子,同时对MYB转录因子家族的基因表达、蛋白保守结构域及系统发育树等内容重点分析。结果红花转录组测序数据中共获得731个转录因子,分属42个转录因子家族。MYB转录因子家族成员的表达及蛋白结构分析显示,MYB转录因子家族在花冠中表达差异显著,氨基酸序列中存在3个保守的蛋白核心结构,系统发育树分析显示聚为2类MYB亚族。结论红花MADs-box、b HLH、MYB、AP2转录因子家族成员最多,在不同组织间表达差异显著,MYB转录因子家族具有一定保守性。
Objective To identify and classify the family of transcription factors in Carthamus tinctorius, and to analyze the differences in the expression levels among different tissues. Methods Unigene was assembled by sequencing data of safflower transcriptome. The transcriptional factor information of Nr database was used to align and annotate safflower transcription factor. Meanwhile, the gene expression, protein conserved domains and phylogenetic tree of MYB transcription factor family were emphasized analysis. Results A total of 731 transcription factors were obtained in the safflower transcriptome sequencing data, belonging to 42 transcription factor families. The MYB transcription factor family members expression and protein structure analysis showed that the MYB transcription factor family in the corolla significant differences in the amino acid sequence of three conserved core protein structure, phylogenetic tree analysis shows that the polyglass 2 MYB subfamily. Conclusions The members of MADs-box, b HLH, MYB and AP2 transcription factors are the most abundant in safflower plants, and the expression of MIBs-box, bFHH, MYB and AP2 genes is significantly different between different tissues. The MYB transcription factor family is conservative.
