目的探讨转化生长因子(transforminggrowthfactor-β1,TGF-β1)诱导的肾小管上皮细胞间质转分化过程中twist表达的改变及意义。方法将人肾小管上皮细胞株(humankidneytubularepithelialcellline,HKCs)细胞分为正常对照组、10ng/mlTGF-β1组;48h后观察细胞形态,逆转录聚合酶链反应(RT-PCR),免疫组织化学测定E-钙黏素(E-cadherin)的表达,免疫印迹测定twist的表达;同时选取不同浓度TGF-β1(0、5ng/ml、10ng/ml、20ng/ml)刺激HKCs细胞,48h后测定twist的表达。结果正常体外培养的HKCs经TGF-β1培养48h后twist蛋白表达明显增加(P<0.05),同时E-cadherin表达明显下降(P<0.05),同时TGF-β1对twist蛋白表达的影响呈浓度依赖性(P<0.05)。结论 TGF-β1诱导肾小管上皮细胞间质转分化过程中存在twist的激活。
Objective To investigate the changes and significance of twist expression in the process of transforming growth factor (TGF-β1) induced by transforming growth factor-β1 (TGF-β1) in the process of stromal transdifferentiation of renal tubular epithelial cells. Methods The human renal tubular epithelial cells (HKCs) were divided into normal control group and 10ng / ml TGF-β1 group. After 48 hours, the cell morphology, reverse transcription polymerase chain reaction (RT-PCR) and immunohistochemistry E The expression of E-cadherin was detected by Western blotting. The expression of twist was determined by Western blotting. HKCs cells were also stimulated with different concentrations of TGF-β1 (0,5ng / ml, 10ng / ml and 20ng / ml) expression. Results The expressions of twist protein were significantly increased (P <0.05) and the expression of E-cadherin in HKCs cultured with TGF-β1 for 48h (P <0.05), and the effect of TGF-β1 on twist protein was concentration-dependent (P <0.05). Conclusion TGF-β1 induces the activation of twist in the process of renal tubular epithelial cell mesenchymal transition.
