目的建立同时测定新疆维药芹菜根提取物中3种黄酮类成分含量的HPLC方法。方法 poroshell EC-C_(18)色谱柱(4.6 mm×150 mm,2.7μm),流动相为乙腈-0.1%磷酸(20∶80),流速0.6 ml·min~(-1),检测波长340 nm,柱温28℃,进样量5μl。结果芹菜苷、3’-甲氧基芹菜苷、芹菜素-7-O-β-D-吡喃葡萄糖苷的质量分别在0.046~0.274μg(r=0.9999),0.025~0.153μg(r=0.9999),0.010~0.062μg(r=0.9998)内与峰面积线性关系良好,加样回收率均值分别为97.81%,97.02%,99.85%,RSD为1.81%,1.50%,1.61%。结论表明所建立的方法简便、快速、准确、重现性好,可用于新疆维药芹菜根提取物中黄酮类成分的含量测定。
OBJECTIVE To establish a HPLC method for simultaneous determination of three flavonoid constituents in celery root extract from Xinjiang Uygur Autonomous Region. Methods The mobile phase consisted of acetonitrile - 0.1% phosphoric acid (20:80) with a flow rate of 0.6 ml · min ~ (-1) and a detection wavelength of 340 nm on a Poroshell EC-C 18 column (4.6 mm × 150 mm, 2.7 μm) , Column temperature 28 ℃, injection volume 5μl. Results The mass of celery glycoside, 3’-methoxy celery glycoside and apigenin-7-O-β-D-glucopyranoside were 0.046 ~ 0.274μg, r = 0.9999, ), 0.010 ~ 0.062μg (r = 0.9998). The average recoveries were 97.81%, 97.02% and 99.85%, respectively. The RSDs were 1.81%, 1.50% and 1.61%, respectively. The results showed that the established method was simple, rapid, accurate and reproducible. It could be used to determine the content of flavonoids in celery root extract of Xinjiang Uygur medicine.
