目的研究环孢素A对小鼠脾细胞不同细胞因子生成的影响及其相互之间的关系。方法利用已建立的Th1细胞反应体系,ELISA法检测在不同浓度环孢素A条件下,BALB/c鼠脾细胞生成IFNγ、IL12及IL4的产量。结果环孢素A抑制IFNγ分泌的最佳浓度为10-1μg/ml,环孢素A抑制IFNγ分泌的同时不影响IL12的产量。高浓度(101μg/ml)环孢素A可促进鼠脾细胞分泌IFNγ和IL12。结论环孢素A对小鼠脾细胞分泌IFNγ有双向调节作用。高浓度环孢素A可促进鼠脾细胞分泌IL12。环孢素A对IFNγ的调节可能与IL12没有直接的关系。
Objective To study the effect of cyclosporin A on the production of different cytokines in mouse spleen cells and their relationship with each other. Methods The established Th1 cell reaction system was used to detect the production of IFNγ, IL12 and IL4 in splenocytes of BALB / c mice under different concentrations of cyclosporine A by ELISA. Results The optimal concentration of cyclosporine A for IFNγ secretion was 10-1μg / ml. Cyclosporine A inhibited the secretion of IFNγ without affecting the production of IL-12. Cyclosporin A at high concentration (101μg / ml) can promote the secretion of IFNγ and IL12 from mouse spleen cells. Conclusion Cyclosporine A can regulate the secretion of IFNγ from mouse spleen cells in a bidirectional manner. High concentrations of cyclosporine A can promote murine spleen cells to secrete IL12. The regulation of IFNγ by cyclosporin A may not have a direct relationship with IL12.
