Dof(DNA-binding one zinc finger)基因家族是植物特有的一类包含高度保守的C_2-C_2锌指结构域的转录因子,其在植物种子发育与萌发、光、激素以及逆境响应中起到重要调控作用。本研究基于已公布的粗山羊草基因组数据,采用生物信息学方法鉴定了粗山羊草Dof基因,并对其结构、保守结构域、染色体定位、系统进化树和表达谱进行了详细的分析。结果表明:粗山羊草Dof转录因子家族包含10个基因,它们均含有完整的C_2-C_2锌指保守结构域;每个基因含有1~3个内含子,AetDof家族蛋白共包含11个保守的模体;该家族基因除7号染色体外,在其余染色体上均有分布;与拟南芥Dof蛋白构建系统发育树发现,该家族基因可被分为3个亚族;转录组数据分析显示,粗山羊草Dof基因的表达具有组织特异性,其中3个转录因子(AetDof1,AetDof7,AetDof10)在种子中特异性强表达。本研究结果为深入解析粗山羊草Dof基因的功能提供了参考依据。
The Dof (DNA-binding one zinc finger) family of genes is a plant-specific transcription factor that contains a highly conserved C 2 -C 2 zinc finger domain that plays an important role in plant seed development and germination, light, hormones, and stress response Regulation. In this study, based on the published genomic data of Aegilops tauschii, the Dof gene was identified by bioinformatics methods and its structure, conserved domain, chromosome location, phylogenetic tree and expression profile were analyzed in detail. The results showed that the Dof transcriptional factor family contained 10 genes, all of which contained a complete C_2-C_2 zinc finger conserved domain. Each gene contained 1 to 3 introns. The AofDof family of proteins contained 11 conserved The gene was distributed on the remaining chromosomes except chromosome 7. The phylogenetic tree of Arabidopsis Dof protein was found to be divided into three subfamilies. According to the transcriptome data analysis, The expression of Dof gene in C. graminearum was tissue-specific, and three transcription factors (AetDof1, AetDof7 and AetDof10) were strongly expressed in seeds. The results of this study provide a reference for further analysis of the function of Dof gene in Aegilops tauschii.
