目的探讨鸟苷酸环化酶C信使核糖核酸(GC-C mRNA)及其配体内生性肽类激素鸟苷素mRNA(GN mRNA)在胃癌组织中的表达及其临床意义。方法应用实时荧光定量PCR法检测GC-C和GN mRNA在60例胃癌和相应30例远癌胃黏膜组织中的表达。结果 GC-C和GNmRNA在胃癌组织中高表达(40.95±25.20和36.95±21.66),而正常胃黏膜组织中不表达。两者在肠型胃癌中的表达高于弥漫型(P<0.01),而与年龄、性别、病灶大小、临床病理分期、分化程度、浸润深度、淋巴结转移和Borrmann分型等因素无关(P>0.05)。两者在胃癌中的表达呈正相关(P<0.01)。结论 GC-C和GN的异常表达与胃黏膜癌变有关,可能参与肠型胃癌的发生发展。
Objective To investigate the expression and clinical significance of guanylate cyclase C messenger RNA (GC-C mRNA) and its ligand endogenous peptide, guanylingenin (GN mRNA) in gastric cancer. Methods The expression of GC-C and GN mRNA was detected by real-time fluorescence quantitative PCR in 60 cases of gastric carcinoma and 30 cases of distant gastric mucosa tissues. Results GC-C and GN mRNA were highly expressed in gastric cancer (40.95 ± 25.20 and 36.95 ± 21.66), but not in normal gastric mucosa. The two expressions were significantly higher in diffuse type (P <0.01) than in diffuse type (P <0.01), but not with age, gender, size of tumor, clinical stage, differentiation, depth of invasion, lymph node metastasis and Borrmann classification (P> 0.05). There was a positive correlation between them in gastric cancer (P <0.01). Conclusion The abnormal expression of GC-C and GN is related to the carcinogenesis of gastric mucosa and may be involved in the development of intestinal-type gastric cancer.
