目的:观察中药有效组分淫羊藿总黄酮、薯蓣皂苷元对围绝经期大鼠内分泌激素水平、卵巢病变等影响。方法:自然衰老围绝经期雌性SD大鼠30只,随机分为3组,各10只,分别是中药低、高剂量组,老年模型组,另设6月龄左右雌性SD大鼠10只为青年对照组。中药组给予中药复方有效成分8周,观察阴道涂片变化,检测大鼠血清雌二醇(E_2)、促性腺激素释放激素(GnRH)、卵泡刺激素(FSH)和黄体生成素(LH)值,免疫蛋白印迹法检测卵巢凋亡蛋白半胱天冬酶(Caspase-3)、血管内皮生长因子(VEGF)蛋白含量变化。结果:与老年模型组比较,青年对照组阴道涂片中有核上皮细胞百分率、E_2水平明显较高,LH、FSH、Caspase-3、VEGF的水平明显较低,差异均有统计学意义(P<0.05,P<0.01);中药高剂量组阴道涂片中有核上皮细胞百分率、E_2、VEGF水平明显升高,Caspase-3的水平明显降低,差异均有统计学意义(P<0.05,P<0.01);中药低剂量组阴道涂片中有核上皮细胞百分率明显升高,差异有统计学意义(P<0.01)。结论:淫羊藿总黄酮、薯蓣皂苷元合用能升高血清E_2和卵巢VEGF水平,降低卵巢Caspase-3的表达,从而延缓大鼠围绝经期的进程。
OBJECTIVE: To observe the effects of Epimedium sagittatum flavonoids and diosgenin on endocrine hormone levels and ovarian lesions in perimenopausal rats. Methods: Thirty female SD rats of spontaneous senescence were randomly divided into 3 groups with 10 rats in each group. They were low dose and high dose of traditional Chinese medicine group and old model group, and 10 female SD rats of 6 months old Youth control group. The traditional Chinese medicine group was given active ingredients of Chinese medicine for 8 weeks to observe the change of vaginal smears. The levels of serum estradiol (E 2), gonadotropin-releasing hormone (GnRH), follicle stimulating hormone (FSH) and luteinizing hormone (LH) The levels of Caspase-3 and Vascular Endothelial Growth Factor (VEGF) in the ovaries were detected by Western blot. Results: Compared with the aged model group, the percentage of nucleated epithelial cells in the vaginal smear of young control group was significantly higher, the level of E_2 was significantly higher, the levels of LH, FSH, Caspase-3 and VEGF were significantly lower, the difference was statistically significant (P <0.05, P <0.01). The percentages of nuclear epithelial cells, E_2 and VEGF in vaginal smears of high-dose group were significantly increased, and the levels of Caspase-3 were significantly decreased (P <0.05, P <0.01). The percentages of nucleated epithelial cells in vaginal smears of low-dose group were significantly increased (P <0.01). Conclusion: Epimedium saponin and diosgenin can increase the levels of serum E_2 and ovarian VEGF, decrease the expression of Caspase-3 in ovary, and delay the process of perimenopause in rats.
