目的研究钾离子通道蛋白KCNJ11基因区的3个标签单核苷酸多态性位点(SNP),及其组成的单体型与糖尿病前期人群运动干预前后糖脂代谢相关指标变化的关系,探讨其作为糖代谢异常人群对运动干预敏感的分子标记。方法对70名糖尿病前期人群进行为期12周的有氧耐力运动干预,测定其干预前后糖脂代谢相关生化指标。采用MALDI-TOF MS方法解析受试者基因组DNA多态位点基因型,分析干预前后相关指标的变化与单个位点及多个位点组成的单体型的相关性。结果 rs5219位点的CC基因型与HOMA-IR、TC、TG指标的下降、与HDL-C指标的升高相关,rs2285676位点的GG基因型与FPG、HbA1c、HOMA-IR、TC的下降相关;rs5219-rs2285676-rs5218位点组成的T-A-G单体型与HOMA-IR的下降相关,C-G-A单体型与TC指标的下降相关,rs5219-rs5218位点组成的C-A单体型与FPG的下降相关,rs2285676-rs5218位点组成的G-G单体型与HDL-C的升高相关。结论KCNJ11基因rs5219位点的CC基因型、rs2285676位点的GG基因型及部分单体型与运动干预前后糖脂代谢的不同指标变化有相关性,可作为评价糖尿病前期人群对运动干预敏感性的参考指标。
Objective To investigate the association of three SNPs in the KCNJ11 gene region and haplotypes of the KCNJ11 gene with the changes of glucose and lipid metabolism in pre-diabetic subjects before and after exercise intervention It is a molecular marker sensitive to exercise intervention in people with abnormal glucose metabolism. Methods A total of 70 pre-diabetic subjects were enrolled in this study. Aerobic endurance exercise intervention was performed for 12 weeks. The biochemical parameters related to glycolipid metabolism before and after intervention were measured. Genomic DNA polymorphism loci genotypes were analyzed by MALDI-TOF MS. The correlations between the changes of related indicators before and after intervention and the haplotypes of single locus and multiple loci were analyzed. Results The reduction of CC genotype and HOMA-IR, TC and TG index at rs5219 locus was correlated with the increase of HDL-C index. The GG genotype of rs2285676 locus correlated with the decrease of FPG, HbA1c, HOMA-IR and TC ; rs5219-rs2285676-rs5218 TAG haplotype was associated with the decline of HOMA-IR, CGA haplotype was associated with the decline of TC, rs5219-rs5218 CA haplotype was associated with the decline of FPG, The GG haplotype of rs2285676-rs5218 was associated with the elevation of HDL-C. Conclusion The CC genotype at rs5219 of KCNJ11 gene and GG genotype and some haplotypes at rs2285676 locus correlate with the changes of different indexes of glycolipid metabolism before and after exercise intervention, which may be used as a measure to evaluate the susceptibility of pre-diabetic patients to exercise intervention reference indicator.
