目的探讨缺氧诱导因子1α(HIF-1α)、碳酸酐酶Ⅸ(CAⅨ)及葡萄糖转运蛋白1(Glut-1)在宫颈癌发生、发展中的作用。方法采用免疫组织化学SP法检测宫颈鳞状细胞癌(宫颈癌组,64例)、宫颈上皮内瘤变(CIN组,57例)和正常宫颈黏膜上皮组织(对照组,26例)中HIF-1α、CAⅨ及Glut-1的表达,分析其与宫颈癌临床病理特征和预后之间的关系。结果宫颈癌组、CIN组和对照组HIF-1α阳性表达率分别为73.44%、43.86%和7.69%(P<0.05),CAⅨ阳性表达率分别为78.13%、54.39%和11.54%(P<0.05),Glut-1阳性表达率分别为51.56%、26.32%和0(P<0.05)。宫颈癌组织中HIF-1α阳性表达与肿瘤组织学分级及临床分期相关(P<0.05);CAⅨ阳性表达与肿瘤组织学分级和淋巴结转移相关(P<0.05);Glut-1阳性表达与肿瘤大小及患者年龄相关(P<0.05)。宫颈癌组织中HIF-1α与CAⅨ的阳性表达呈正相关(r=0.400,P<0.05),HIF-1α与Glut-1的阳性表达呈正相关(r=0.693,P<0.05)。HIF-1α与CAⅨ阳性表达宫颈癌患者的生存率低于阴性表达者(P<0.05)。结论HIF-1α、CAⅨ和Glut-1蛋白在宫颈癌组织中均呈过表达;HIF-1α蛋白的阳性表达在宫颈癌诊断及分期中有一定价值;宫颈癌组织中HIF-1α和CAⅨ蛋白阳性表达与患者较差的预后密切相关。
Objective To investigate the role of HIF-1α, CAⅨ and Glut-1 in the development of cervical cancer. Methods Immunohistochemical SP method was used to detect the expression of HIF-1α in cervical squamous cell carcinoma (cervical cancer group, 64 cases), cervical intraepithelial neoplasia (CIN group, 57 cases) and normal cervical epithelial tissue (control group, 26 cases) 1α, CAⅨ and Glut-1 in cervical cancer and to analyze the relationship between them and the clinicopathological features and prognosis of cervical cancer. Results The positive expression rates of HIF-1α in cervical cancer group, CIN group and control group were 73.44%, 43.86% and 7.69%, respectively (P <0.05). The positive expression rates of CAⅨ were 78.13%, 54.39% and 11.54% ), Glut-1 positive rates were 51.56%, 26.32% and 0 (P <0.05) respectively. The positive expression of HIF-1α in cervical cancer was correlated with histological grade and clinical stage (P <0.05). The positive expression of CAⅨ was correlated with tumor grade and lymph node metastasis (P <0.05). The positive expression of Glut-1 was positively correlated with tumor size And patient age (P <0.05). The positive expression of HIF-1alpha and CAIX in cervical cancer tissues was positively correlated (r = 0.400, P <0.05). The positive expression of HIF-1alpha and Glut-1 was positively correlated (r = 0.693, P <0.05). The survival rates of HIF-1α and CAⅨ-positive cervical cancer patients were lower than those of negative ones (P <0.05). Conclusion The expression of HIF-1α, CAⅨ and Glut-1 protein in cervical cancer tissues was overexpressed. The positive expression of HIF-1α protein had some value in the diagnosis and staging of cervical cancer.
