目的:探讨Toll样受体(Toll-like receptors,TLR)4和TLR9在胃癌组织中的表达及其与临床病理特征的关系。方法:收集2009年1月至2009年10月在解放军第174医院手术切除的42例胃癌及其癌旁(距癌灶边缘5 cm)组织标本。RT-PCR和Western blotting检测胃癌组织中TLR4、TLR9在mRNA和蛋白水平的表达。结果:胃癌组织中TLR4和TLR9 mRNA表达高于癌旁组织(1.29±0.03 vs 0.53±0.02,0.99±0.04 vs 0.22±0.05;均P<0.05),Ⅲ期+Ⅳ期胃癌组织中TLR4和TLR9mRNA表达显著高于Ⅰ期+Ⅱ期(P<0.05),有远处转移的胃癌组织中TLR4和TLR9 mRNA表达显著高于无远处转移者(P<0.05);TLR4和TLR9 mRNA的表达水平与患者的年龄、性别、淋巴结转移没有相关性。胃癌组织中TLR4和TLR9蛋白的表达高于癌旁组织(1.36±0.05 vs 0.48±0.04,1.12±0.05 vs 0.34±0.03;均P<0.05),Ⅲ期+Ⅳ期胃癌组织TLR4和TLR9蛋白的表达显著高于Ⅰ期+Ⅱ期(P<0.05)。结论:TLR4和TLR9在胃癌组织中高表达,其表达与胃癌分期、远处转移具有相关性,表明TLR4和TLR9可能参与了胃癌的发生和发展,可能为胃癌的治疗提供一种潜在的靶点。
Objective: To investigate the expression of Toll-like receptors (TLR) 4 and TLR9 in gastric carcinoma and its relationship with clinicopathological features. Methods: Twenty-four gastric cancer tissues and their adjacent tissues (5 cm away from the edge of the tumor) surgically removed from the People’s Liberation Army No.174 Hospital from January 2009 to October 2009 were collected. The mRNA and protein expression of TLR4 and TLR9 in gastric cancer tissues were detected by RT-PCR and Western blotting. Results: The expressions of TLR4 and TLR9 mRNA in gastric cancer tissues were significantly higher than those in paracancerous tissues (1.29 ± 0.03 vs 0.53 ± 0.02, 0.99 ± 0.04 vs 0.22 ± 0.05; all P <0.05) (P <0.05). The expressions of TLR4 and TLR9 mRNA in gastric cancer with distant metastasis were significantly higher than those without distant metastasis (P <0.05). The expressions of TLR4 and TLR9 mRNA in patients with distant metastasis The age, gender, lymph node metastasis were not related. The expressions of TLR4 and TLR9 in gastric cancer tissues were significantly higher than those in paracancerous tissues (1.36 ± 0.05 vs 0.48 ± 0.04, 1.12 ± 0.05 vs 0.34 ± 0.03; all P <0.05). The expressions of TLR4 and TLR9 in stage Ⅲ + Which was significantly higher than that in stage Ⅰ and Ⅱ (P <0.05). Conclusion: TLR4 and TLR9 are highly expressed in gastric cancer tissues. The expression of TLR4 and TLR9 is correlated with staging and distant metastasis of gastric cancer, indicating that TLR4 and TLR9 may be involved in the occurrence and development of gastric cancer and may provide a potential target for the treatment of gastric cancer.
