目的:探讨大鼠肝纤维化不同分期肝表观扩散系数(apparent diffusion coefficient,ADC)与基质金属蛋白酶抑制剂-1(tissue inhibitor of metalloproteinase-1,TIMP-1)mRNA表达的相关性。方法:采用猪血清腹腔注射联合高脂饮食建立肝纤维化大鼠模型。给药4周后开始,取实验组大鼠48只和对照组大鼠12只行磁共振弥散加权成像(diffusion weighted imaging,DWI)检查,计算b值=800 s/mm~2时的ADC值。DWI检查后快速处死大鼠,行病理检查,采用RT-PCR检测TIMP-1 mRNA表达。成模大鼠按肝纤维化病理分期结果又分为S0期组(n=4)、S1期组(n=11)、S2期组(n=12)、S3期组(n=10)和S4期组(n=9),比较肝纤维化各组ADC值及TIMP-1 mRNA的表达,并分析两者的相关性。结果:对照组、肝纤维化S1~4期组大鼠肝ADC值及TIMP-1 mRNA表达差异有统计学意义(分别F=46.54和53.87,均P<0.05),ADC值除对照组与肝纤维化S1期组、S1期组与S2期组、S2期组与S3期组比较差异无统计学意义(均P>0.05)外,其余各组间比较均有统计学意义(均P<0.05);各组TIMP-1 mRNA除肝纤维化S1期组与S2期组、S3期组与S4期组比较差异无统计学意义(均P>0.05)外,其余各组间差异均有统计学意义(均P<0.05)。秩相关分析显示肝ADC值与TIMP-1 mRNA表达的变化呈负相关(r=–0.76,P<0.01)。结论:随大鼠肝纤维化进程不断加深,ADC值逐渐减低,TIMP-1 mRNA表达逐渐升高,二者呈负相关。
Objective: To investigate the relationship between the apparent diffusion coefficient (ADC) and the expression of tissue inhibitor of metalloproteinase-1 (TIMP-1) mRNA in different stages of hepatic fibrosis in rats. Methods: The rat model of hepatic fibrosis was established by intraperitoneal injection of pig serum combined with high-fat diet. Four weeks after the start of administration, 48 rats in the experimental group and 12 rats in the control group were examined by diffusion weighted imaging (DWI), and the ADC value at b = 800 s / mm ~ 2 was calculated . Rats were sacrificed immediately after DWI and pathological examination was performed. The expression of TIMP-1 mRNA was detected by RT-PCR. The model rats were divided into S0 group (n = 4), S1 group (n = 11), S2 group (n = 12), S3 group S4 group (n = 9). The ADC value and TIMP-1 mRNA expression in each group were compared, and the correlation between them was analyzed. Results: The liver ADC value and TIMP-1 mRNA expression in the control group and the liver fibrosis stage S1 ~ 4 group were significantly different (F = 46.54 and 53.87, P <0.05, respectively) There was no significant difference between S1 group, S1 group and S2 group, S2 group and S3 group (all P> 0.05), the other groups were statistically significant (all P <0.05 ); There was no significant difference in the expression of TIMP-1 mRNA among the groups except S1 group and S2 group, S3 group and S4 group (all P> 0.05) Significance (all P <0.05). Rank correlation analysis showed that liver ADC value was negatively correlated with the change of TIMP-1 mRNA expression (r = -0.76, P <0.01). CONCLUSION: With the progression of hepatic fibrosis in rats, the ADC value gradually decreases and the expression of TIMP-1 mRNA gradually increases, both of which are negatively correlated.
