目的 探讨细胞凋亡和细胞周期相关基因蛋白与经典型霍奇金淋巴瘤临床分期的关系。方法 应用免疫组化ABC法检测bcl 2 ,ABC和TUNEL双标记技术检测细胞凋亡 ,ABC和SABAP双标记方法检测MIB1、TopoⅡα、Rb和 p2 1在 6 2例经典型霍奇金淋巴瘤组织芯片中的表达。结果 6 2例经典型霍奇金淋巴瘤中 35例bcl 2阳性 ,在 6 2例非肿瘤性背景细胞中均有凋亡小体 ,而在H RS细胞仅有 10例胞质内见凋亡小体 ;H RS细胞bcl 2表达与细胞凋亡呈负相关性 (P <0 0 5 ) ;MIB1及TopoⅡα高表达与临床分期呈正相关性 (P <0 0 1) ,反之 ,Rb及p2 1高表达与临床分期呈负相关性 (P <0 0 1)。所有抗原的表达与霍奇金淋巴瘤的组织学亚型均无相关性 (P >0 0 5 )。结论 bcl 2的过度表达可以阻断H RS细胞的凋亡 ;CD30阳性H RS细胞中MIB1及TopoⅡα表达指数随病程的进展而升高 ,而Rb及 p2 1表达指数随病程的进展而降低 ,他们可作为霍奇金淋巴瘤预后判断的参考指标。霍奇金淋巴瘤的病程进展可能与细胞凋亡和细胞周期相关基因及蛋白表达的变化有关
Objective To investigate the relationship between apoptosis and cell cycle related protein and clinical stage of Hodgkin’s lymphoma. Methods The expressions of bcl-2, ABC and TUNEL were detected by immunohistochemical ABC method. MIB1, TopoⅡα, Rb and p21 were detected by ABC and SABAP double labeling in 62 cases of Hodgkin lymphoma tissue microarray In the expression. Results Of the 62 classic Hodgkin ’s lymphoma cases, 35 were bcl - 2 positive and apoptotic bodies were present in 62 non - tumorous background cells, while only 10 in H RS cells showed cytoplasmic apoptosis The expression of bcl-2 in HRS cells was negatively correlated with cell apoptosis (P <0.05). The positive expression of MIB1 and TopoⅡα were positively correlated with clinical stage (P <0.01), while Rb and p21 High expression was negatively correlated with clinical stage (P <0.01). There was no correlation between the expression of all the antigens and histological subtypes of Hodgkin’s lymphoma (P> 0.05). Conclusion The overexpression of bcl 2 can block the apoptosis of H RS cells. The expression index of MIB1 and Topo Ⅱ α in CD30 positive H RS cells increases with the progression of the disease, while the expression indices of Rb and p2 1 decrease with the progression of the disease. Hodgkin lymphoma can be used as a prognostic indicator of reference. The progress of Hodgkin’s lymphoma may be related to the changes of apoptosis and cell cycle related gene and protein expression
