目的:观察三氧化二砷体外对神经母细胞瘤细胞增殖的影响及其作用机制的探讨。方法:①MTT法观察三氧化二砷对神经母细胞瘤细胞生长的影响;②化学染色观察实验和对照组细胞形态学改变;③PI染色后流式细胞仪检测实验和对照组细胞凋亡峰;④凝胶电泳观察凋亡细胞DNA的“梯型”条带。结果:①三氧化二砷明显抑制神经母细胞瘤细胞株SJNSH细胞增殖,抑制作用呈剂量和作用时间依赖性;②通过细胞形态学、流式细胞仪对DNA的测定及DNA凝胶电泳均证实,三氧化二砷可诱导SJNSH细胞发生凋亡。在一定浓渡范围内,凋亡率与三氧化二砷的浓度和作用时间呈依赖关系。结论:三氧化二砷可明显抑制神经母细胞瘤细胞增殖,其作用机制主要是通过诱导肿瘤细胞发生调亡
Objective: To observe the effect of arsenic trioxide on the proliferation of neuroblastoma cells in vitro and its mechanism of action. METHODS: 1MTT method was used to observe the effect of arsenic trioxide on the growth of neuroblastoma cells; 2 chemical staining was observed in experimental and control groups; morphological changes were observed after 3 PI staining; apoptosis peaks were detected by flow cytometry and in the control group;4 gel electrophoresis; Observe the “ladder” band of apoptotic DNA. RESULTS: 1Arsenic trioxide significantly inhibited the proliferation of neuroblastoma cell line SJ-N SH cells, and the inhibitory effect was dose- and time-dependent; 2 The DNA quantification by flow cytometry and DNA gel electrophoresis were both used. It was confirmed that arsenic trioxide induced apoptosis in SJ-N-SH cells. Within a certain concentration range, the apoptotic rate is dependent on the concentration and time of action of arsenic trioxide. Conclusion: Arsenic trioxide can significantly inhibit the proliferation of neuroblastoma cells. Its mechanism is mainly through the induction of apoptosis of tumor cells.
