为了研究云南不同地区烟草内生细菌的多样性及分布特征。在全省4个不同区域采集相同烟草品种33个样品,通过分离培养烟草内生细菌、16S r DNA序列分析获得不同烟区烟草内生细菌的多样性特征。从不同地区烟草叶片中共分离得到内生细菌372株,分别归属于11个属。以Bacillus属为优势菌群,已鉴定出7个种,包括B.pumilus,B.amyloliquefaciens,B.cereus,B.subtilis,B.flexus,B.simplex,B.megaterium;B.pumilus和Pantoea agglomerans类群细菌是4个地区共同的优势菌群;香农多样性指数由大到小依次为文山、大理、昭通、玉溪;其中玉溪和大理地区样品的相似性系数最高,为0.58,玉溪和昭通地区样品的相似性系统最低,为0.31。烟草地理分布对烟草内生细菌组成具有一定的影响,烟草内生细菌的种类多样性特征在不同地区间存在差异,部分内生细菌的分布存在地域专一性。研究结果为认识烟草内生细菌的特征及挖掘利用有益内生细菌奠定了基础,并为内生细菌在其它植物中的研究提供理论依据。
In order to study the diversity and distribution characteristics of tobacco endophytic bacteria in different regions of Yunnan. 33 samples of the same tobacco variety were collected from 4 different areas of the province. The endophytic bacteria in different tobacco growing areas were obtained by isolating and cultivating tobacco endophytic bacteria and 16S r DNA sequence analysis. 372 endophytic bacteria were isolated from tobacco leaves in different regions, belonging to 11 genera. Seven species are identified with the genus Bacillus as the dominant species, including B. pumilus, B. amyloliquefaciens, B. cereus, B. subtilis, B. flexus, B. simplex, B. megaterium; B. pumilus and Pantoea agglomerans The group of bacteria is the common dominant microflora in the four regions. The Shannon-Dyer diversity index is Wenshan, Dali, Zhaotong and Yuxi in descending order. The similarity coefficient of Yuxi and Dali samples is the highest (0.58) The lowest similarity system is 0.31. The geographical distribution of tobacco has certain influence on the composition of tobacco endophytic bacteria. The species diversity of tobacco endophytic bacteria is different in different regions, and the distribution of some endophytic bacteria is region-specific. The results provide a theoretical basis for understanding the characteristics of tobacco endophytic bacteria and excavating and utilizing beneficial endophytic bacteria, and providing a theoretical basis for the study of endophytic bacteria in other plants.
