目的探讨羟基磷灰石(HAP)纳米粒子体外抗白血病作用。方法采用MTT法检测单用HAP和联合化疗药物对K562细胞的增殖抑制及化疗增敏作用,并用倒置显微镜观察药物对K562细胞形态学的影响。结果①HAP和抗癌药物对K562细胞增殖均可产生影响,在倒置显微镜下,可见全部测定孔细胞增殖均较对照孔低下。②K562细胞生长抑制率与HAP的浓度和作用时间呈明显正相关。③HAP与化疗药物合用具有更良好的抑癌效果。结论HAP纳米粒子明显抑制K562细胞生长,具有体外抗白血病作用和化疗增敏作用;HAP纳米粒子对白血病及肿瘤的治疗将具有良好的开发应用前景。
Objective To investigate the anti-leukemic effect of hydroxyapatite (HAP) nanoparticles in vitro. Methods MTT assay was used to detect the proliferation inhibition and chemosensitivity of K562 cells treated with HAP alone and combined chemotherapy. The morphological changes of K562 cells were observed by inverted microscope. Results ① Both HAP and anti-cancer drugs could affect the proliferation of K562 cells. Under inverted microscope, all the measured cell proliferation was lower than that of the control wells. ②K562 cell growth inhibition rate and HAP concentration and duration of action was positively correlated. ③ HAP combined with chemotherapy drugs have a better anti-cancer effect. Conclusion HAP nanoparticle significantly inhibits the growth of K562 cells and has the anti-leukemia effect and chemosensitization effect in vitro. The treatment of leukemia and tumor by HAP nanoparticles will have a good prospect of development and application.
