目的探讨荧光定量PCR(FQ-PCR)技术检测耐甲氧西林金黄色葡萄球菌(MRSA)的临床作用。方法选取从医院ICU病人分离出的32株金黄色葡萄球菌(金葡菌)和22株非金黄色葡萄球菌。利用荧光PCR技术和传统检测方法分离培养进行检测,对检测结果不符的菌株最终进行测序鉴定,依据鉴定效果进行比较,确定FQ-PCR法的特异性及灵敏度。结果 FQ-PCR与传统培养法对金葡菌检测准确率为100.00%,均测出32株阳性。FQ-PCR与培养药敏试验检测MRSA的符合率为93.75%,而培养药敏试验法检测为阴性而PCR法检测为阳性的2株经最后确定为阳性。结论 FQ-PCR法可用于临床对MRSA的实时检测,具有较高的临床诊断价值。
Objective To investigate the clinical application of fluorescence quantitative PCR (FQ-PCR) in detecting methicillin-resistant Staphylococcus aureus (MRSA). Methods 32 strains of Staphylococcus aureus (Staphylococcus aureus) and 22 strains of Staphylococcus aureus isolated from hospital ICU patients were selected. Fluorescent PCR technology and traditional detection methods were used for isolation and culture. The strains that did not match the test results were sequenced and compared with each other to determine the specificity and sensitivity of FQ-PCR. Results The accuracy of FQ-PCR and traditional culture method was 100.00% for Staphylococcus aureus, and 32 were positive. The coincidence rate of FQ-PCR and culture sensitivity test for detection of MRSA was 93.75%, while the two strains tested negative by culture sensitivity test and positive by PCR test were finally confirmed as positive. Conclusion FQ-PCR method can be used in clinical real-time detection of MRSA, with high clinical diagnostic value.
