目的进行药用植物桃儿七Sinopodophyllum hexandrum的遗传多样性研究,为桃儿七资源的保护和开发利用提供分子遗传理论基础。方法对6个桃儿七居群进行DALP分析,利用PopGene 1.31软件分析遗传多样性相关数据,UPGMA方法聚类,结合Treeview软件生成树状图。结果筛选5对引物组合,扩增共产生150条DNA片段,其中104条谱带具有遗传多态性,占69.33%,平均每组引物扩增所得多态条带为20.8,6个居群平均多态百分率为14.22%。6居群总的观察等位基因数Na为1.693 3,平均1.142 2;总的有效等位基因数Ne为1.417 1,平均1.083 8;基因多样性指数H为0.244 3,平均0.048 8;总的Shannon多样性指数I为0.364 3,平均0.073 0;居群总的基因多样性Ht为0.244 3,居群内基因多样性Hs为0.048 7。基因分化系数Gst为0.800 5,即遗传变异有80.05%发生在居群间,19.95%发生在居群内,居群每代迁移数Nm为0.124 6。结论桃儿七居群间存在较大的遗传分化,基因流受到阻碍,这与桃儿七的繁育系统和生存环境紧密相关。
Objective To study the genetic diversity of Sinopodophyllum hexandrum, a medicinal plant, and provide the molecular genetic basis for the conservation and utilization of Taoyu seven resources. Methods DALP analysis was carried out on seven populations of six peach populations. PopGene 1.31 software was used to analyze the genetic diversity related data, UPGMA method was clustered, and Treeview software was used to generate dendrogram. Results A total of 150 DNA fragments were screened by PCR amplification, of which 104 bands had genetic polymorphism (69.33%). The average number of polymorphic bands amplified by each primer set was 20.8, and the average of 6 populations Polymorphic percentage was 14.22%. The total number of alleles observed in 6 populations was 1.693 3 with an average of 1.142 2. The total number of effective alleles, Ne, was 1.417 1 with an average of 1.083 8. The genetic diversity index H was 0.244 3 with an average of 0.048 8. Shannon’s diversity index I was 0.364 3, with an average of 0.073 0; the total genetic diversity of the population Ht was 0.244 3, and the genetic diversity within the population Hs was 0.048 7. The coefficient of genetic differentiation, Gst, was 0.800 5, which means that 80.05% of the genetic variation occurred among the populations and 19.95% occurred within the population. The population migration per generation was 0.124 6. Conclusion There is a large genetic differentiation among the populations of Taoyuan, and the gene flow is hindered, which is closely related to the breeding system and living environment of Taoshi seven.
