本文报道用反相高效液相色谱法测定484例服用卡马西平、苯妥英和苯巴比妥的癫痫病人的血清药物浓度。采用UltraspheieODS(250×4.6mm,5μm)色谱柱,0.01mol/L磷酸盐缓冲液(PH2.5)/甲醇(45:55)为流动相,洗脱经乙酸乙酯提取的血清样品,在254nm处检测药物和内标的峰高,以内标法定量,并对方法的回收率、准确度和精密度进行评价。血药浓度监测结果提示:三种药物在剂量不变时,浓度均波动较大,故动态监测抗癫痫药物的血药浓度对临床合理用药具有重要意义。
This article reports the determination of serum drug concentrations in 484 epileptic patients taking carbamazepine, phenytoin, and phenobarbital using reversed-phase high performance liquid chromatography. The serum samples extracted with ethyl acetate were eluted with Ultraspheie ODS (250 × 4.6 mm, 5 μm) column and 0.01 mol / L phosphate buffer (PH 2.5) / methanol (45:55) as the mobile phase. The peak height of the drug and internal standard was measured at 254 nm and quantified by internal standard method, and the recovery, accuracy and precision of the method were evaluated. Blood concentration monitoring results suggest that: the three drugs at constant dose, the concentration fluctuations are large, so the dynamic monitoring of antiepileptic drugs plasma concentration of clinical rational use of drugs is of great significance.
