目的 :研究三氧化二砷 (As2 O3)抑制肝癌细胞增殖、诱导肝癌细胞凋亡以及对骨髓抑制等作用。方法 :建立肝癌裸鼠移植瘤模型 ,应用 TU NEL 法观察不同浓度的 As2 O3对裸鼠移植瘤凋亡指数 (AI)的影响 ;应用免疫组化 SABC法检测不同浓度的 As2 O3对增殖细胞核抗原阳性细胞指数 (PI)的影响 ;检测移植瘤裸鼠的 WBC并进行骨髓涂片检查。结果 :经 As2 O3治疗的移植瘤裸鼠的骨髓有核细胞增生活跃 ,粒红比例正常 ,巨核细胞多见 ;低、中浓度 As2 O3治疗裸鼠移植瘤的 AI高于对照组 (P <0 .0 1和 P <0 .0 5 ) ;高浓度组 AI与对照组比较 ,无显著性差异 (P >0 .0 5 )。低、中浓度组 PI低于对照组 (P <0 .0 1) ;高浓度组 PI与对照组比较 ,无显著性差异 (P >0 .0 5 )。结论 :As2 O3通过抑制肿瘤细胞增殖、诱导细胞凋亡而达到其抗肝癌的效果 ,As2 O3对移植瘤裸鼠骨髓无抑制作用。
Objective: To study the effects of arsenic trioxide (As2O3) on the proliferation of hepatoma cells, induction of apoptosis of hepatocellular carcinoma cells and myelosuppression. Methods: The nude mice transplanted tumor model of hepatocellular carcinoma was established. The effect of different concentrations of As2O3 on apoptosis index (AI) of transplanted tumors in nude mice was observed by TU NEL method; As2O3 with different concentrations was used to detect proliferating cell nuclear antigen by immunohistochemical SABC method. The effect of positive cell index (PI); detection of WBC in xenografted nude mice and bone marrow smear. RESULTS: The proliferation of bone marrow nucleated cells was normal in transplanted nude mice treated with As2O3. The proportion of erythroblasts was normal, megakaryocytes were more common, and the AI of low and medium concentration As2O3 in nude mice was higher than that in the control group (P < 0). .0 1 and P <0.05.) There was no significant difference between the high-dose AI group and the control group (P > 0.05). The PI in the low and medium concentration groups was lower than that in the control group (P < 0.01). There was no significant difference in the PI between the high concentration group and the control group (P > 0.05). Conclusion : As2O3 can achieve anti-hepatocarcinoma effect by inhibiting tumor cell proliferation and inducing cell apoptosis. As2O3 has no inhibitory effect on bone marrow of transplanted tumor nude mice.
