目的检测乳腺癌组织中死亡相关蛋白激酶(DAPK)的表达,探讨DAPK在乳腺癌发生、发展中的作用。方法采用免疫组化SP法检测42例乳腺增生、30例乳腺不典型增生、14例乳腺非浸润性癌及68例乳腺浸润性癌的DAPK表达水平。结果乳腺浸润性癌组织中DAPK的阳性表达率为42.6%,明显低于乳腺增生(92.9%)、乳腺不典型增生(73.3%)、乳腺非浸润性癌组织中的表达(71.4%,均为P<0.05);非浸润性癌和不典型增生组织中的DAPK阳性表达率为72.7%,亦低于乳腺增生组织(P<0.05),但是非浸润性癌和不典型增生两组间无显著性差异(P>0.05)。研究还发现DAPK阳性表达与乳腺癌的雌激素受体的表达、淋巴结转移、病理分级、组织分型等相关。结论DAPK表达降低对乳腺癌的演化和进展具有一定重要作用,可用于乳癌的早期诊断及靶向治疗。
Objective To detect the expression of death-associated protein kinase (DAPK) in breast cancer and to explore the role of DAPK in the development and progression of breast cancer. Methods The immunohistochemical SP method was used to detect the expression of DAPK in 42 cases of breast hyperplasia, 30 cases of atypical hyperplasia of breast, 14 cases of non-invasive breast carcinoma and 68 cases of invasive breast carcinoma. Results The positive rate of DAPK expression in invasive breast cancer was 42.6%, which was significantly lower than that in hyperplasia of mammary gland (92.9%), atypical hyperplasia of mammary gland (73.3%) and non-invasive breast cancer (71.4% P <0.05). The positive rate of DAPK in non-invasive carcinoma and atypical hyperplasia was 72.7%, which was also lower than that in mammary gland hyperplasia (P <0.05), but there was no significant difference between non-invasive carcinoma and atypical hyperplasia Sex differences (P> 0.05). The study also found that the positive expression of DAPK and breast cancer estrogen receptor expression, lymph node metastasis, pathological grading, tissue typing and so on. Conclusion The decrease of DAPK expression plays an important role in the development and progression of breast cancer. It can be used in early diagnosis and targeted therapy of breast cancer.
