目的探讨青蒿酸体外作用对人肝癌细胞SMMC-7721细胞增殖的影响及其机制。方法采用MTT法检测青蒿酸对SMMC-7721细胞增殖的影响,Hoechst-33258、Annexin V-FITC/PI检测细胞凋亡情况。结果青蒿酸作用SMMC-7721细胞24,48,72 h,能显著抑制细胞增殖,并呈时间、剂量依赖性。青蒿酸作用48 h后,SMMC-7721呈明显的凋亡形态,高浓度青蒿酸作用48 h后,呈典型的细胞凋亡特征,凋亡率达67.54%。结论青蒿酸能抑制SMMC-7721细胞增殖,这可能和诱导细胞凋亡有关。
Objective To investigate the effect of artemisinic acid on the proliferation of human hepatoma SMMC-7721 cells in vitro and its mechanism. Methods The effect of artemisinic acid on the proliferation of SMMC-7721 cells was detected by MTT assay. The apoptosis of SMMC-7721 cells was detected by Hoechst-33258 and Annexin V-FITC / PI. Results Artemisinic acid SMMC-7721 cells 24,48,72 h, can significantly inhibit cell proliferation in a time-and dose-dependent manner. At 48 h, the apoptosis of SMMC-7721 cells was obvious, and the apoptotic rate of SMMC-7721 cells was 67.54% after treated with high concentration of artemisinic acid for 48 h. Conclusion Artemisinic acid can inhibit the proliferation of SMMC-7721 cells, which may be related to the induction of apoptosis.
