目的在表达绿色荧光蛋白(green fluorescent protein,GFP)裸小鼠脑内建立人脑多形性胶质母细胞瘤(glioblastoma multiforme,GBM)原位移植模型。方法将表达GFP的C57/B6鼠与Balb/c nu/nu系裸小鼠杂交筛选出表达GFP的裸小鼠。把人脑GBM组织原位移植于GFP裸小鼠脑内,荧光显微镜下观察移植瘤的生长特性、血管生成及表皮生长因子受体(epidermal growth factor receptor,EGFR)表达。结果成功培育出表达GFP的荧光裸小鼠;移植瘤中EGFR阳性表达率(78.6±5.2)%,肿瘤侵袭性生长并血管丰富。结论人脑GBM荧光裸小鼠原位移植模型能模拟人脑原发恶性胶质瘤高侵袭及EGFR高表达的生物学特性。
Objective To establish a glioblastoma multiforme (GBM) orthotopic transplantation model in nude mice expressing green fluorescent protein (GFP). Methods C57 / B6 mice expressing GFP and Balb / c nu / nu mice were screened to express GFP-expressing nude mice. Human GBM tissues were transplanted in situ into the brain of GFP nude mice, and the growth characteristics, angiogenesis and expression of epidermal growth factor receptor (EGFR) were observed under a fluorescence microscope. Results Fluorescent nude mice expressing GFP were successfully established. The positive rate of EGFR in the transplanted tumor was 78.6 ± 5.2%. The tumor grew aggressive and had abundant blood vessels. Conclusion The in situ orthotopic transplantation of GBM in nude mice can mimic the biological characteristics of high invasiveness of human primary malignant glioma and high EGFR expression.
