摘要 采用磷酸钙-DNA共沉淀法,将IL-2真核表达载体直接注射至经大剂量化疗后的小鼠腹腔内,可成功地转染腹腔巨噬细胞(MФ),并表达基因产物,能刺激脾脏淋巴细胞增殖,提高腹腔MФ的细胞毒活性,并促使腹腔MФ产生大量的IL-1、TNF和NO。还能使化疗后处于低水平的白细胞和血小板显著回升,降低化疗后白细胞和血小板数量的减少程度,并显著提高外周血淋巴细胞和单核细胞的数量。提示IL-2基因转染MФ可能有助于肿瘤患者放疗、化疗后造血功能损伤的恢复和免疫功能重建。
Abstract By using calcium phosphate-DNA co-precipitation method, IL-2 eukaryotic expression vector was injected directly into the abdominal cavity of mice after high-dose chemotherapy, and peritoneal macrophages (MФ) can be successfully transfected, and gene products are expressed. It can stimulate the proliferation of spleen lymphocytes, increase the cytotoxic activity of abdominal MФ, and promote the production of large amounts of IL-1, TNF and NO in abdominal cavity MФ. It can also significantly increase leukocytes and platelets at low levels after chemotherapy, reduce the reduction in the number of white blood cells and platelets after chemotherapy, and significantly increase the number of peripheral blood lymphocytes and monocytes. It is suggested that the transfection of IL-2 gene into MФ may contribute to the recovery of hematopoietic function and the reconstruction of immune function after radiotherapy and chemotherapy.
