目的观察重组人内皮抑素(rhES)对小鼠肺腺癌LA795生长和转移的抑制作用。方法对ThES高效表达克隆pCX的表达产物进行纯化,得到ThES。将LA795肺腺癌细胞接种于T739小鼠背部皮下,随机分成2组,接种后第10天起分别给予rhES20mg/kg·b.w.或等体积PBS缓冲液皮下注射,1次/d,共14d。观察2组肿瘤生长情况、肺湿重、肺表面转移结节数、动物生存时间,分别进行t检验。结果治疗前后rhES组肿瘤体积分别为(650±201)mm3、(1642±211)mm3;而PBS组肿瘤体积由治疗前的(623±248)mm3增至(9194±952)mm3。rhES组肺湿重、肺表面转移结节数明显少于PBS组,动物生存时间明显延长(P<0.01)。结论rhES可明显抑制LA795所致的小鼠实验性肿瘤的生长与转移,延长动物的生存时间。
Objective To observe the inhibitory effect of recombinant human endostatin (rhES) on the growth and metastasis of mouse lung adenocarcinoma LA795. Methods The expression products of ThCs were cloned and purified to obtain ThES. LA795 lung adenocarcinoma cells were inoculated subcutaneously into the back of T739 mice and randomly divided into 2 groups. RhES 20 mg/kg b.w. or equal volume of PBS buffer was injected subcutaneously once a day from 10 days after inoculation, once a day for 14 days. The tumor growth, lung wet weight, number of lung metastatic nodules, and survival time of the animals in the two groups were observed and t-tests were performed. Results The tumor volume in the rhES group was (650±201) mm3 and (1642±211) mm3 before and after treatment, while the tumor volume in the PBS group increased from (623±248) mm3 before treatment to (9194±952) mm3. The wet weight of lung and the number of metastatic nodules on the lung surface in the rhES group were significantly less than those in the PBS group, and the survival time of the animals was significantly longer (P<0.01). Conclusion rhES can significantly inhibit the growth and metastasis of experimental tumors induced by LA795 and prolong the survival time of animals.
