目的:探讨胃泌素类似物五肽胃泌素(PG)及胃泌素受体拮抗剂丙谷胺(PGL)对体外培养的人结肠癌细胞株HT29生长的影响。方法:采用MTT法检测细胞增殖情况用流式细胞术(FCM)分析细胞周期变化。结果:高浓度的PG(10-5~10-7M)能明显促进HT29细胞的增殖(P<005),低浓度的PG(10-8~10-10M)对HT29细胞无明显促增殖作用(P>005)。PGL能抑制PG对HT29细胞的刺激作用,并对HT29细胞的增殖有直接抑制作用(P<005)。细胞周期分析显示PG能缩短细胞G3/G1期,PGL则延长细胞G0/G1期(P<005)。结论:胃泌素对体外培养的HT29细胞的生长具有调节作用,胃泌素受体拮抗剂可能在结肠癌的治疗中发挥重要作用。
Objective: To investigate the effect of gastrin analog pentagastrin (PG) and gastrin receptor antagonist proglumide (PGL) on the growth of human colon cancer cell line HT29 in vitro. Methods: Cell proliferation was measured by MTT assay. Cell cycle changes were analyzed by flow cytometry (FCM). Results: High concentration of PG (10-5~10-7M) can significantly promote the proliferation of HT29 cells (P<0.05). Low concentration of PG (10-8~10-10M) does not significantly promote proliferation of HT29 cells. Role (P>005). PGL can inhibit the stimulatory effect of PG on HT29 cells and directly inhibit the proliferation of HT29 cells (P<005). Cell cycle analysis showed that PG could shorten the G3/G1 phase, and PGL prolonged G0/G1 phase (P<005). Conclusion: Gastrin can regulate the growth of HT29 cells cultured in vitro. Gastrin receptor antagonists may play an important role in the treatment of colon cancer.
