槐耳颗粒对体外培养的人卵巢癌SKOV-3细胞增殖和凋亡的影响

目的 :观察槐耳颗粒对卵巢癌SKOV-3细胞增殖和凋亡的影响,并探讨其可能的作用机制。方法 :取对数生长期的SKOV-3细胞,用不同质量浓度的槐耳颗粒(0、2、4和8 mg/m L)处理48 h后,在倒置相差显微镜下观察SKOV-3细胞形态的变化;CCK-8法检测槐耳颗粒对SKOV-3细胞增殖的影响;FCM法检测槐耳颗粒对SKOV-3细胞凋亡的影响;蛋白质印迹法检测槐耳颗粒对SKOV-3细胞中凋亡相关蛋白Bcl-2、Bax和cleaved-caspase 3蛋白表达的影响。结果 :用不同质量浓度的槐耳颗粒处理SKOV-3细胞48 h后倒置显微镜下观察发现,各组SKOV-3细胞的形态均发生改变。CCK-8检测结果显示,不同质量浓度的槐耳颗粒能显著抑制SKOV-3细胞的增殖(P<0.05),且呈时间和剂量依赖性;槐耳颗粒能诱导SKOV-3细胞发生凋亡,与对照组相比,早期凋亡率明显增加,差异有统计学意义(P<0.05);蛋白质印迹法检测结果显示,槐耳颗粒可下调Bcl-2蛋白的表达水平(P<0.05),而上调Bax和cleaved-caspase3蛋白的表达水平(P值均<0.05)。结论 :槐耳颗粒在体外可抑制人卵巢癌SKOV-3细胞增殖并诱导其凋亡。 Objective: To observe the effects of Huaier granules on proliferation and apoptosis of ovarian cancer SKOV-3 cells and to explore its possible mechanism. Methods: SKOV-3 cells in logarithmic growth phase were treated with different concentrations of Huaier particles (0, 2, 4 and 8 mg / m L) for 48 h. The morphology of SKOV-3 cells was observed under inverted phase contrast microscope The effects of Huaier granules on the proliferation of SKOV-3 cells were detected by CCK-8 assay. The apoptosis of SKOV-3 cells was detected by FCM assay. The apoptosis of SKOV-3 cells was detected by Western blotting Effects of Bcl-2, Bax and cleaved-caspase 3 proteins on the expression of the related proteins. Results: The SKOV-3 cells were treated with different concentrations of Huaier particles 48 hours after inverted microscope and found that the morphology of SKOV-3 cells in each group were changed. CCK-8 test results showed that different concentrations of Huaier particles can significantly inhibit the proliferation of SKOV-3 cells (P <0.05), and in a time and dose-dependent manner; Huaier particles can induce apoptosis of SKOV-3 cells, Compared with the control group, the early apoptotic rate increased significantly (P <0.05). Western blotting showed that Huaier granule could down-regulate the expression of Bcl-2 protein (P <0.05) Up-regulated the expression of Bax and cleaved-caspase3 protein (all P <0.05). Conclusion: Huaier granule can inhibit the proliferation and induce the apoptosis of human ovarian cancer SKOV-3 cells in vitro.
